The dams are limnic ecosystems of great importance for its multiple uses, among them, water supply for the public and to culture of artisanal fish are most relevant. The aim of the present study is to evaluate the physical-chemical characteristics and the phytoplankton community in two chosen sites (Point 1 littoral zone of point source; Point 2 pelagic zone of non-point source) of the Minister João Alves dam, which is also known as Boqueirão de Parelhas/RN. This represents the spatial distribution of the phytoplankton species in order to understand any possible alterations of the water quality and the phytoplankton composition in relation to the water quality originating from the impact of the tilapia, Oreochromis niloticus, culture. The study period also encompasses temporal variations exhibited in two seasons of an annual cycle, one during the dry season (Oct, Nov and Dec of 2008 and Jan of 2009), and the other rainy season (Mar, Apr, May and June of 2008) to extend the observation. The physicalchemical parameters, such as pH, temperature, electrical conductivity, concentration of dissolved oxygen were measured in situ and the values of the inorganic nutrients (nitrate, ammonium and orto-phosfato) and chlorophyll in the laboratory. The quali-quantitative analyses of the phytoplankton had been carried through sedimentation technique and the enumeration of the random of 400 cells, colonies and filaments counted using Sedgwick-Rafter counting chamber. The results of pH varied widely from the acidic to alkaline range with the minimum of 5.8 (± 0.8) and the maximum of 9.2 (± 0.7-0.8), at point 1 and 2. The dissolved oxygen content was higher in the rainy period than that in the dry period. The maximum electrical conductivity was of 1409 μScm-1 in point 1 and 431 minim of μScm-1, in point 2. There was a considerable alteration in the levels of inorganic nutrients such as nitrate-nitrogen, ammoniacal nitrogen and orthophosphate during the two cycles of study period. Phytoplankton assemblages presented a picture of alternate dominance among species Cyanobacteria, Bacillariophyceae and Chlorophyceae. The trophic state index diagnosed to the category of mesotrophic, which is based on the values of chlorophyll, total phosphorus and Secchi-disc measurements. The wind driven turbulence of the water column and the fresh inflow of water (flushing and dilution) during rainy season acted as constraint and did-not allow an exaggerated growth of the species of cyanobacteria. On the basis of the present we conclude that the culture of tilapias in cage-culture fails to produce pollution load that could compromise the quality of the water of the dam, probably be due to small dimension of the culture in relation to the size, volume of the water and the reservoir capacity support its own environment

En el presente trabajo se muestran los resultados experimentales obtenidos del cultivo del consorcio de microalgas (Chlorella sp y Scenedesmus sp.) empleando el efluente urbano pretratado proveniente de la planta de tratamiento cloacal ―La Viñita‖, Provincia de Catamarca, Argentina. Los objetivos se orientan a caracterizar el consorcio de microalgas autóctonas en cuanto a la relación de la producción de biomasa y lípidos, en condiciones controladas de cultivo, en efluentes urbanos provenientes de la planta de tratamiento de líquidos cloacales ―La Viñita‖ y analizar la eficiencia del proceso biorremediador realizado por las microalgas, considerando la eficiencia de los tratamientos, en relación a los parámetros que corresponden a días de cultivo, estado del consorcio y propiedades físico-químicas del sobrenadante. Se emplearon tres variables (% de Inóculo, % de medio nutricional e inyección de CO2), el ensayo se realizó a lo largo de 12 días de cultivo ―indoor‖. Se analizaron los datos de producción de biomasa en cada tratamiento, se determinó la concentración de lípidos presentes en la biomasa y se hizo observación microscópica de los distintos tratamientos al finalizar el ensayo y un seguimiento macroscópico durante el ensayo. Se eligieron los tratamientos eficientes en términos de biomasa y lípidos. En todos los casos se empleó fotoperiodo 12 hs/12 hs, sin control de esterilidad y con agitación manual. Sobre cada muestra se realizaron las determinaciones de biomasa, lípidos y pH. Considerando los aspectos de desarrollo de las microalgas y los parámetros físico-químicos, los resultados obtenidos indicaron que los tratamientos con inyección de CO2 alcanzaron mayor productividad de biomasa algal y lípidos. En las condiciones de 20% de inóculo, inyección de CO2, empleo de efluente con refuerzo de medio nutricional se obtiene en 3 días: 58 mg/L de biomasa y la producción más alta del ensayo en cuanto a lípidos (16,8 mg/L). En cuanto al proceso biorremediador se observó que disminuyó la concentración de DBO5 en un 63,63 %, la de DQO en un 51,8 %, la de nitratos en un 38 % y aumentó la cantidad de fosfato, lo que podrá ser corregido con modificaciones en las concentraciones de sales en el medio de cultivo. El efluente resultante no presenta riesgo de sodicidad por lo que podría ser utilizada para riego. Se observó un cambio de color (de verde claro a verde oscuro) y se confirmó con la observación microscópica la predominancia de Chlorella sp. en un 90 %, la presencia de rotíferos en baja cantidad y ausencia de otras microlagas no deseadas como filamentosas y diatomeas. No se detectó la presencia de huevo de helmintos.

As microalgas podem ser consideradas como um dos mais eficientes sistemas biológicos de transformação de energia solar em compostos orgânicos. Quando cultivadas em meios adequados, certas espécies podem duplicar sua biomassa diariamente. Além disso, possuem inúmeras vantagens, como: elevada velocidade de crescimento; potencial para absorver CO2, reduzindo assim a quantidade de emissões deste gás na atmosfera e diminuindo o efeito estufa. O objetivo do trabalho foi estudar o efeito do uso de pentoses no cultivo de Chlorella minutissima, Chlorella vulgaris, Chlorella homosphaera, Dunaliella salina, Spirulina paracas e Synechococcus nidulans, avaliando o perfil cinético do crescimento e a capacidade de produção de carboidratos e proteínas. Para o cultivo das microalgas foram utilizados os meios: Zarrouk, Bristol`S Modificado e DUN. Em todos os meios o componente nitrogenado foi reduzido pela metade e utilizado 1%, 5%, 10%, 20% e 30% de pentoses, com concentrações de xilose e arabinose que representassem as mesmas presentes em caldo hidrolisado do bagaço de cana de açúcar pré-tratado. Os cultivos foram realizados em fotobiorreatores de 2 L, mantidos em estufa a 30 ºC, fotoperíodo de 12h claro/escuro e 2500 Lx, com agitação a uma vazão de 0,75 v.v.m. . O crescimento de biomassa foi monitorado diariamente pela densidade ótica das culturas em espectrofotômetro a 670nm. Foram avaliados parâmetros cinéticos como a concentração máxima de biomassa, produtividade máxima e velocidade específica máxima de crescimento. A determinação do consumo das pentoses foi realizada através da metodologia de Somogy e Nelson, para a determinação de carboidratos foi utilizada uma adaptação do método do ácido 3,5 dinitro salicílico, as proteínas foram quantificadas pelo método de micro-Kjeldahl. Todas as microalgas foram capazes de consumir em no máximo quatro dias as concentrações de pentoses, e logo após esta etapa mixotrófica manter-se em crescimento autotrófico, destacando-se as cepas de Dunaliella salina e Synechococcus nidulans que esgotaram as maiores concentrações utililizadas em dois dias de cultivo. Para as cianobactérias estudadas, Spirulina paracas cultivada com 10% de C5, foi a que obteve os melhores resultados de concentração celular, produtividade e velocidade específica de crescimento máxima, 1,364 g.L-1 , 0,128 g.L-1 .dia-1 e 0,240 dia-1 . Em relação ao efeito na composição da biomassa, Synechococcus nidulans produziu o maior teor de proteínas, 62,9%, nos ensaios com 10% de C5. Já as cepas de Chlorophytas os melhores resultados foram obtidos com o uso de 5% de C5, para os parâmetros cinéticos destacam-se os valores encontrados para Dunaliella salina, onde a maior concentração de biomassa, produtividade e velocidade específica de crescimento foram 1,246 g.L-1 , 0,091 g.L- 1 .dia-1 e 0,379 dia-1 , respectivamente. Chlorella minutissima e Dunaliella salina foram as melhores produtoras de carboidratos, alcançando 58,6%/0,3 g.L-1 e 23,07%/0,29 g.L-1 ,respecivamente. Logo, o uso de pentoses nas microalgas em substituição as fontes tradicionais de carbono, resultou no crescimento das mesmas, o que mostra que estas podem agir como intermediários para a absorção de açúcares de cinco carbonos.

O trabalho teve por objetivo avaliar a dinâmica de nitrogênio, em cultivo heterotrófico, a partir da cianobactéria Aphanothece microscopica Nägeli, sob o escopo de uma biorrefinaria. Neste sentido, foi avaliada a contribuição dos compostos nitrogenados não proteicos, na dinâmica de distribuição do nitrogênio, na biomassa gerada pelo micro-organismo em estudo, quando cultivado em sistema autotrófico e heterotrófico. Para o cultivo em condições autotróficas, foi utilizado o meio padrão BG-11, enquanto que, para o cultivo em condições heterotróficas, foi empregado o efluente da indústria de laticínios. Inicialmente, foi avaliada a contribuição dos pigmentos na fração nitrogenada não proteica tendo como base dois experimentos. No primeiro experimento foi selecionada a melhor condição para a produção de pigmentos, expressos pela clorofila-a em sistema heterotrófico, tendo como base os parâmetros C/N (20, 40 e 60), N/P (5, 10 e 15) e concentração de inóculo (100, 200 e 300 mg.L-1), mediante um planejamento fatorial 23 . Os experimentos foram conduzidos em biorreator heterotrófico a 20°C, pH 7,6 e aeração contínua de 1VVM. A melhor condição de produção de pigmento foi indicada como sendo a 200 mg.L-1 de concentração celular, razões C/N 20 e N/P 10. Com base nestes resultados, um segundo experimento foi delineado, visando avaliar a contribuição de pigmentos na fração de nitrogênio não proteico, bem como avaliar a produção de clorofila-a e ficobiliproteínas (ficocianina, aloficocianina e ficoeritrina), sob influência da luz e do meio de cultivo. Foi possível destacar teores superiores de ficobiliproteínas na biomassa gerada no cultivo heterotrófico. No entanto, com notada diferença (p≤0,05) nos teores de clorofila-a, quando são comparadas as concentrações na biomassa de meios autotróficos (10,7 mg.g-1) e heterotróficos (1,0 mg.g-1). Fato este compensado pelo menor tempo de cultivo registrado para atingir o final do experimento, quando o micro-organismo é cultivado em condições heterotróficas. Fica demonstrado assim, ainda, a importante contribuição dos pigmentos na fração de nitrogênio não proteico. Na sequência, um terceiro e quarto experimentos foram delineados, visando avaliar a influência do nitrogênio inorgânico intracelular na fração não proteica e na produção de proteína, assim como a caracterização da fração proteica quanto ao seu perfil aminoacídico. O estudo da dinâmica do nitrogênio intracelular demonstrou que o N-NH4 + foi a forma nitrogenada predominante, perfazendo importante fração de N-NP, sendo, portanto, os teores de N-NP significativamente dependente dos teores de pigmentos e nitrogênio intracelular. Os aminogramas das biomassas geradas pelos cultivos autotróficos e heterotróficos indicaram como aminoácidos majoritários o ácido glutâmico e aspártico, seguidos por valina, leucina e isoleucina, e como minoritários, lisina, glicina e metionina. O perfil aminoacídico caracterizou-se por apresentar aminoácidos essenciais como isoleucina, metionina + cisteína, fenilalanina + tirosina, valina e treonina em concentrações superiores ao preconizado pela FAO/WHO. A caracterização da fração proteica quanto ao perfil aminoacídico qualificou esta biomassa como fonte potencial de proteína. Os resultados obtidos neste trabalho demonstram a influência e dinâmica de distribuição dos compostos nitrogenados em Aphanothece microscopica Nägeli. Fica demonstrado, ainda, que a implementação do conceito de biorrefino, no tipo de agroindústria estudado, poderá representar importantes possibilidades de aproveitamento sustentável do efluente gerado.

A oportunidade de produção de biomassa microalgal tem despertado interesse pelos diversos destinos que a mesma pode ter, seja na produção de bioenergia, como fonte de alimento ou servindo como produto da biofixação de dióxido de carbono. Em geral, a produção em larga escala de cianobactérias e microalgas é feita com acompanhamento através de análises físicoquímicas offline. Neste contexto, o objetivo deste trabalho foi monitorar a concentração celular em fotobiorreator raceway para produção de biomassa microalgal usando técnicas de aquisição digital de dados e controle de processos, pela aquisição de dados inline de iluminância, concentração de biomassa, temperatura e pH. Para tal fim foi necessário construir sensor baseado em software capaz de determinar a concentração de biomassa microalgal a partir de medidas ópticas de intensidade de radiação monocromática espalhada e desenvolver modelo matemático para a produção da biomassa microalgal no microcontrolador, utilizando algoritmo de computação natural no ajuste do modelo. Foi projetado, construído e testado durante cultivos de Spirulina sp. LEB 18, em escala piloto outdoor, um sistema autônomo de registro de informações advindas do cultivo. Foi testado um sensor de concentração de biomassa baseado na medição da radiação passante. Em uma segunda etapa foi concebido, construído e testado um sensor óptico de concentração de biomassa de Spirulina sp. LEB 18 baseado na medição da intensidade da radiação que sofre espalhamento pela suspensão da cianobactéria, em experimento no laboratório, sob condições controladas de luminosidade, temperatura e fluxo de suspensão de biomassa. A partir das medidas de espalhamento da radiação luminosa, foi construído um sistema de inferência neurofuzzy, que serve como um sensor por software da concentração de biomassa em cultivo. Por fim, a partir das concentrações de biomassa de cultivo, ao longo do tempo, foi prospectado o uso da plataforma Arduino na modelagem empírica da cinética de crescimento, usando a Equação de Verhulst. As medidas realizadas no sensor óptico baseado na medida da intensidade da radiação monocromática passante através da suspensão, usado em condições outdoor, apresentaram baixa correlação entre a concentração de biomassa e a radiação, mesmo para concentrações abaixo de 0,6 g/L. Quando da investigação do espalhamento óptico pela suspensão do cultivo, para os ângulos de 45º e 90º a radiação monocromática em 530 nm apresentou um comportamento linear crescente com a concentração, apresentando coeficiente de determinação, nos dois casos, 0,95. Foi possível construir um sensor de concentração de biomassa baseado em software, usando as informações combinadas de intensidade de radiação espalhada nos ângulos de 45º e 135º com coeficiente de determinação de 0,99. É factível realizar simultaneamente a determinação inline de variáveis do processo de cultivo de Spirulina e a modelagem cinética empírica do crescimento do micro-organismo através da equação de Verhulst, em microcontrolador Arduino.

The roles of weather variability and sunspots in the occurrence of cyanobacteria blooms, were investigated using cyanobacteria cell data collected from the Fred Haigh Dam, Queensland, Australia. Time series generalized linear model and classification and regression (CART) model were used in the analysis. Data on notified cell numbers of cyanobacteria and weather variables over the periods 2001 and 2005 were provided by the Australian Department of Natural Resources and Water, and Australian Bureau of Meteorology, respectively. The results indicate that monthly minimum temperature (relative risk [RR]: 1.13, 95% confidence interval [CI]: 1.02-1.25) and rainfall (RR: 1.11; 95% CI: 1.03-1.20) had a positive association, but relative humidity (RR: 0.94; 95% CI: 0.91-0.98) and wind speed (RR:0.90; 95% CI: 0.82-0.98) were negatively associated with the cyanobacterial numbers, after adjustment for seasonality and auto-correlation. The CART model showed that the cyanobacteria numbers were best described by an interaction between minimum temperature, relative humidity, and sunspot numbers. When minimum temperature exceeded 18%C and relative humidity was under 66%, the number of cyanobacterial cells rose by 2.15-fold. We conclude that the weather variability and sunspot activity may affect cyanobacterial blooms in dams.

In June 2011 a large phytoplankton bloom resulted in a catastrophic mortality event that affected a large coastal embayment in the Solomon Islands. This consisted of an area in excess of 20 km2 of reef and soft sandy habitats in Marovo Lagoon, the largest double barrier lagoon in the world. This embayment is home to over 1200 people leading largely subsistence lifestyles depending on the impacted reefs for majority of their protein needs. A toxic diatom (Psuedo-nitzchia spp.) and toxic dinoflagellate (Pyrodinium bahamense var. compressum) reached concentrations of millions of cells per litre. The senescent phytoplankton bloom led to complete de-oxygenation of the water column that reportedly caused substantial mortality of marine animal life in the immediate area within a rapid timeframe (24 h). Groups affected included holothurians, crabs and reef and pelagic fish species. Dolphins, reptiles and birds were also found dead within the area, indicating algal toxin accumulation in the food chain. Deep reefs and sediments, whilst initially unaffected, have now been blanketed in large cyanobacterial mats which have negatively impacted live coral cover especially within the deep reef zone (> 6 m depth). Reef recovery within the deep zone has been extremely slow and may indicate an alternative state for the system.

Fish collected after a mass mortality at an artificial lake in south-east Queensland, Australia, were examined for the presence of nodularin as the lake had earlier been affected by a Nodularia bloom. Methanol extracts of muscle, liver, peritoneal and stomach contents were analysed by HPLC and tandem mass spectrometry; histological examination was conducted on livers from captured mullet. Livers of sea mullet (Mugil cephalus) involved in the fish kill contained high concentrations of nodularin (median 43.6 mg/kg, range 40.8-47.8 mg/kg dry weight; n = 3) and the toxin was also present in muscle tissue (median 44.0 mu g/kg, range 32.3-56.8 mu g/kg dry weight). Livers of fish occupying higher trophic levels accumulated much lower concentrations. Mullet captured from the lake 10 months later were also found to have high hepatic nodularin levels. DNA sequencing of mullet specimens revealed two species inhabiting the study lake: M. cephalus and an unidentified mugilid. The two mullet species appear to differ in their exposure and/or uptake of nodularin, with M. cephalus demonstrating higher tissue concentrations. The feeding ecology of mullet would appear to explain the unusual capacity of these fish to concentrate nodularin in their livers; these findings may have public health implications for mullet fisheries and aquaculture production where toxic cyanobacteria blooms affect source waters. This report incorporates a systematic review of the literature on nodularin measured in edible fish, shellfish and crustaceans.

Cyanobacterial mass occurrences, also known as water blooms, have been associated with adverse health effects of both humans and animals. They can also be a burden to drinking water treatment facilities. Risk assessments of the blooms have generally focused on the cyanobacteria themselves and their toxins. However, heterotrophic bacteria thriving among cyanobacteria may also be responsible for many of the adverse health effects, but their role as the etiological agents of these health problems is poorly known. In addition, studies on the water purification efficiency of operating water treatment plants during cyanobacterial mass occurrences in their water sources are rare. In the present study, over 600 heterotrophic bacterial strains were isolated from natural freshwater, brackish water or from treated drinking water. The sampling sites were selected as having frequent cyanobacterial occurrences in the water bodies or in the water sources of the drinking water treatment plants. In addition, samples were taken from sites where cyanobacterial water blooms were surmised to have caused human health problems. The isolated strains represented bacteria from 57 different genera of the Gamma-, Alpha- or Betaproteobacteria, Actinobacteria, Flavobacteria, Sphingobacteria, Bacilli and Deinococci classes, based on their partial 16S rRNA sequences. Several isolates had no close relatives among previously isolated bacteria or cloned 16S rRNA genes of uncultivated bacteria. The results show that water blooms are associated with a diverse community of cultivable heterotrophic bacteria. Chosen subsets of the isolated strains were analysed for features such as their virulence gene content and possible effect on cyanobacterial growth. Of the putatively pathogenic haemolytic strains isolated in the study, the majority represented the genus Aeromonas. Therefore, the Aeromonas spp. strains isolated from water samples associated with adverse health effects were screened for the virulence gene types encoding for enterotoxins (ast, alt and act/aerA/hlyA), flagellin subunits (flaA/flaB), lipase (lip/pla/lipH3/alp-1) and elastase (ahyB) by PCR. The majority (90%) of the Aeromonas strains included one or more of the six screened Aeromonas virulence gene types. The most common gene type was act, which was present in 77% of the strains. The fla, ahyB and lip genes were present in 30 37% of the strains. The prevalence of the virulence genes implies that the Aeromonas may be a factor in some of the cyanobacterial associated health problems. Of the 183 isolated bacterial strains that were studied for possible effects on cyanobacterial growth, the majority (60%) either enhanced or inhibited growth of cyanobacteria. In most cases, they enhanced the growth, which implies mutualistic interactions. The results indicate that the heterotrophic bacteria have a role in the rise and fall of the cyanobacterial water blooms. The genetic and phenotypic characteristics and the ability to degrade cyanobacterial hepatotoxins of 13 previously isolated Betaproteobacteria strains, were also studied. The strains originated from Finnish lakes with frequent cyanobacterial occurrence. Tested strains degraded microcystins -LR and -YR and nodularin. The strains could not be assigned to any described bacterial genus or species based on their genetic or phenotypic features. On the basis of their characteristics a new genus and species Paucibacter toxinivorans was proposed for them. The water purification efficiency of the drinking water treatment processes during cyanobacterial water bloom in water source was assessed at an operating surface water treatment plant. Large phytoplankton, cyanobacterial hepatotoxins, endotoxins and cultivable heterotrophic bacteria were efficiently reduced to low concentrations, often below the detection limits. In contrast, small planktonic cells, including also possible bacterial cells, regularly passed though the water treatment. The passing cells may contribute to biofilm formation within the water distribution system, and therefore lower the obtained drinking water quality. The bacterial strains of this study offer a rich source of isolated strains for examining interactions between cyanobacteria and the heterotrophic bacteria associated with them. The degraders of cyanobacterial hepatotoxins could perhaps be utilized to assist the removal of the hepatotoxins during water treatment, whereas inhibitors of cyanobacterial growth might be useful in controlling cyanobacterial water blooms. The putative pathogenicity of the strains suggests that the health risk assessment of the cyanobacterial blooms should also cover the heterotrophic bacteria.

Currently, the classification used for cyanobacteria is based mainly on morphology. In many cases the classification is known to be incongruent with the phylogeny of cyanobacteria. The evaluation of this classification is complicated by the fact that numerous strains are only described morphologically and have not been isolated. Moreover, the phenotype of many cyanobacterial strains alters during prolonged laboratory cultivation. In this thesis, cyanobacterial strains were isolated from lakes (mainly Lake Tuusulanjärvi) and both morphology and phylogeny of the isolates were investigated. The cyanobacterial community composition in Lake Tuusulanjärvi was followed for two years in order to relate the success of cyanobacterial phenotypes and genotypes to environmental conditions. In addition, molecular biological methods were compared with traditional microscopic enumeration and their ability and usefulness in describing the cyanobacterial diversity was evaluated. The Anabaena, Aphanizomenon, and Trichormus strains were genetically heterogeneous and polyphyletic. The phylogenetic relationships of the heterocytous cyanobacteria were not congruent with their classification. In contrast to heterocytous cyanobacteria, the phylogenetic relationships of the Snowella and Woronichinia strains, which had not been studied before this thesis, reflected the morphology of strains and followed their current classification. The Snowella strains formed a monophyletic cluster, which was most closely related to the Woronichinia strain. In addition, a new cluster of thin, filamentous cyanobacterial strains identified as Limnothrix redekei was revealed. This cluster was not closely related to any other known cyanobacteria. The cyanobacterial community composition in Lake Tuusulanjärvi was studied with molecular methods [denaturant gradient gel electrophoresis (DGGE) and cloning of the 16S rRNA gene], through enumerations of cyanobacteria under microscope, and by strain isolations. Microcystis, Anabaena/Aphanizomenon, and Synechococcus were the major groups in the cyanobacterial community in Lake Tuusulanjärvi during the two-year monitoring period. These groups showed seasonal succession, and their success was related to different environmental conditions. The major groups of the cyanobacterial community were detected by all used methods. However, cloning gave higher estimates than microscopy for the proportions of heterocytous cyanobacteria and Synechococcus. The differences were probably caused by the high 16S rRNA gene copy numbers in heterotrophic cyanobacteria and by problems in the identification and detection of unicellular cyanobacteria.
